Details:
Volumetric Muscle Loss (VML) is a debilitating condition resulting from the loss of a large portion of muscle. Current treatments are ineffective in restoring tissue function due to poor understanding of cellular/molecular processes governing VML repair. Macrophages are crucial in VML outcome; however, much is still unknown about their phenotype, kinetics, and crosstalk with other cells.

To further our knowledge in the underlying mechanism of VML fibrosis in order to design an effective therapeutic strategy, this thesis has been divided into three specific aims: 1) Investigate immune response in Regenerative vs. Fibrotic VML injuries. 2) Elucidate specific cellular mechanisms dictating VML repair and pathology. 3) Improve tissue function via implantation of tunable hydrogel containing encapsulated dexamethasone treated macrophages.

This work will elucidate cellular and molecular mechanism never investigated in VML, particularly regarding the immune system. Such finding will allow for the development of a therapeutic strategy which takes into consideration such mechanisms to achieve therapeutic outcomes never achieved before.
